In molecular biology, G-quadruplex secondary structures (G4) are formed in nucleic acids by sequences that are rich in guanine.[2] They are helical in shape and contain guanine tetrads that can form from one,[3] two[4] or four strands.[5] The unimolecular forms often occur naturally near the ends of the chromosomes, better known as the telomeric regions, and in transcriptional regulatory regions of multiple genes, both in microbes[6][7] and across vertebrates [8][7] including oncogenes in humans.[9] Four guanine bases can associate through Hoogsteen hydrogen bonding to form a square planar structure called a guanine tetrad (G-tetrad or G-quartet), and two or more guanine tetrads (from G-tracts, continuous runs of guanine) can stack on top of each other to form a G-quadruplex.

The placement and bonding to form G-quadruplexes is not random and serve very unusual functional purposes. The quadruplex structure is further stabilized by the presence of a cation, especially potassium, which sits in a central channel between each pair of tetrads.[3] They can be formed of DNARNALNA, and PNA, and may be intramolecularbimolecular, or tetramolecular.[10] Depending on the direction of the strands or parts of a strand that form the tetrads, structures may be described as parallel or antiparallel. G-quadruplex structures can be computationally predicted from DNA or RNA sequence motifs,[11][12] but their actual structures can be quite varied within and between the motifs, which can number over 100,000 per genome. Their activities in basic genetic processes are an active area of research in telomere, gene regulation, and functional genomics research.[13][14]

Those genetic bases (the little flat molecules with one or two rings and lots of “N” and “O” in them) can not only bond to each other in PAIRS between different bases – the base guanine (G) can bond to itself if it makes a circle big enough for FOUR of them instead of just a pair (see left picture above). Sometimes a metal ion in the center can help this happen.

This competing form of binding disrupts the nice, neat, normal “double helix” binding, but not permanently.

When G4 binding happens, there are many working ways that the little squares can form. These squares tend to STACK just like in normal DNA, and so sequences that are rich in guanine can build these stacks easily, if the guanines are in the right places. Alternatively, two or more strands of DNA or RNA can join in these knots, but at a minimum, a single strand can do it, as shown on the right of the graphic above, where the sequence knots up against itself.

Note that this is more like “knots in velcro”, and not like various circling knots we are familiar with.

Why it’s important for COVID vaccines

The COVID vaccine mRNA is not natural – and not only in that normal bases are substituted with modified bases. It also has been “codon optimized”, meaning the very sequence of bases was changed from the viral sequence, by substituting base triplets with synonymous ones, that are known to process faster in humans. But the problem is that these changes are rich in guanine, making the knotting more likely. This not-fully-equivalent substitution has potential pharmacological consequences. In a sense, viral evolutionary intelligence was ignored (a beautiful point made by Kevin McKernan).

MOAR DNA and RNA STUFF

Check out this video, which shows TRANSCRIPTION – the normal, natural way that DNA is turned into RNA in living organisms on this planet.

What you are seeing here is “good”!

NEXT – Somebody else posted this under their own timeline on X.

NOW – the molecular biologist who found the DNA contamination problem in the mRNA vaccines – Kevin McKernan – reposted the latter video with a comment about something which is NOT in the video, but which would change it. Here is his comment.

Let me explain what Kevin is saying.

Kevin is not commenting about the transcription process shown here. He is NOT talking about the red DNA strand, either. He is commenting about the yellow strand of mRNA which is being produced.

“Now add N1 methyl Pseudouridine and watch that yellow strand turn into quadruplex G knot.”

He is saying that if you change the uridines in that molecule to N1-methyl-pseudouridine, as was done in the Pfizer vaccine, the yellow strand of mRNA will “kink up” with G-quadruplex knots.

He explains that further, when somebody wrongly claims the video is showing translation (creation of proteins) and not transcription (creation of mRNA). Observe.

So to sum up, he is saying that the “good” “yellow” mRNA produced in the video was corrupted by the modifications that were made to it by Pfizer.

He says the following things:

  • it’s transcription, not translation, that is being shown
  • the “pseudo-uridine” changes done to the viral mRNA sequence make the mod-RNA dissociate more easily
  • the same changes make the mod-RNA form G4-quadruplexes easier
  • Pfizer also changed the code to include more guanines (G), which means more quadruplexes
  • all the G4-based KNOTS that are present in the mod-RNA cause the subsequent TRANSLATION step (looks similar to the video, but it’s PROTEIN being formed) to suddenly PAUSE, just like a KNOT in a climbing rope will lock up against a device until it is untangled. This is the “ribosomal pausing” he’s talking about.

So how does the DeMarinis paper figure into this?

The DeMarinis paper showed that the Pfizer vaccine can be reverse-transcribed from mod-RNA back into DNA by human liver cells. Reverse transcription would look pretty much like the video of transcription, except that RNA would be read, and DNA would spit out.

Whether that DNA is incorporated into the genome or not is unknown, but it is a huge concern, because another group (Jaenisch paper) showed that the COVID virus SARS-CoV-2 can and does incorporate its spike into the genome of some patients, AND yet another group (Mehedi paper) showed that the viral spike shepherds its own mRNA into the nucleus of human cells, where reverse transcription could happen.

I wanted people to see a bigger point – that this repeated and coincidentally connected “bungling” by Pfizer and its allies, which weirdly matches WEF’s dream of “hacking humanity”, is worthy of a bit of suspicion.

When a trail of disconnected yet matching footprints goes in some direction, it’s rather logical that somebody was purposefully passing that way.

It sure looks to me like the various “negligent” yet clearly convenient aspects of this whole exercise, add up to an attempt to gain a population-wide hack into the human cell nucleus.

Just sayin’!

In my case, Some of the thefts were likely from a pretty close friend who grew up in poverty. Since some cash was taken from a place that was very well concealed there were only a couple of people who would even have known to look there. Also there was a time when she had a turquoise ring that looked Exactly like one I’d gotten on a trip to AZ that I think she lifted from my house at a teenage sleepover.

I never “proved” that these incidents involved her but that’s what my gut was telling me.

I had an expensive textbook lifted from the cafeteria coatrack. I tried to keep my books looking fairly pristine so didn’t mark all along the page edges, like some people did, to make them standout as mine, so perhaps that made it tempting to someone in “need”…

I think Christians are sinful creatures, like everybody is, but we just know that our sins are covered & forgiven by Jesus & His shed blood. Though this process Should be transformative & lead to Believers becoming more Christlike that’s not always how it works, especially with those who come to Christ for some type of “fire insurance”…

My cousin is going back to his mom’s place in January to help out, after my very clever uncle’s passing.

This could be interesting. He may have to take the house apart to matchsticks to be sure he’s recovered everything. He remembers helping his dad move away from California and there were silver ingots stashed in the HVAC ductwork.

Forty years in between them. Charlotte -> Carson city was less than that. (they didn’t run at the same time though.) Charlotte and Dahlonega were casualties of the Civil War, though their accompanying gold fields were in decline before that.

The Confederacy tried to run New Orleans for a while but simply never had the actual specie to do their own coinage. (There was no issue using other countries’ precious metal coinage…but that all disappeared quickly. Gresham’s Law.) That mint was useful enough for the union to resume (and besides, it did silver as well as gold) after the civil war. I believe New Orleans was operational until 1904.

I was in the ammo department at Wallyworld a number of years ago (before the great Obola ammo crunch), and the clerk, noticing I had my eye on a brand of ammo I was curious about (because it was unfamiliar to me) asked me what I shot. I told him CZ-75 and he said don’t shoot that in that gun. I asked what sort of gun would be appropriate he said, “Lorcin. Jennings. High Point.” It was steel case and apparently somewhat corrosive.

This stuff was imported from Eastern Europe…REALLY sketchy stuff.

I believe most of the US manufacturers use non-corrosive primers even in their cheapass stuff.

I’m reminded of an ad I saw one time. Couple of old guys at the range. First one takes a shot with his revolver. Low and left, almost off the page. His buddy looks at him quizzically. They don’t bugger up, and they fit the hand. (Well, I had one that in its old age had trouble with the cheap federal champion stuff from Walmart.) (I see more 1911s malfunction…and the owners all have amnesia after it happens.)

I haven’t followed things the past few years, but there is a full size CZ-75, a compact, and the RAMI subcompact (all interchangeable mags as long as the mag isn’t too short for the gun in question). I tell glock people that the full size is a CZ-17, the compact is a CZ-19 and the RAMI is a CZ-26. That lets them relate things to the common Glock 9mms.

CZ has more recently come out with the “Omega” trigger which is much simpler than the old style, and the guns actually went down significantly in price. Between a sale and a member price, I was able to get a CZ-75B Omega for just under 400 bucks. For an all-metal gun that’s not from a dodgy manufacturer that’s a raging STEAL.

So why worry about the full burden of proof thing, when the preponderance of evidence is what the public is going to take note of.

It makes a distinction between different types of blessings, mentioning liturgical blessings for unions vs. spontaneous blessings given for the reasons listed in the comment.

I understand the distinction, but I wonder how many who receive such a blessing will think This is because I recognize myself as destitute and in need of help. I don’t know if the idea of a need for repentance would be inherent in receiving a blessing from a priest. I think it more likely that they will think, as the comment states, that they “beg that all that is true, good, and humanly valid in their lives and their relationships be enriched, healed, and elevated by the presence of the Holy Spirit.” The implication is that there are good and humanly valid elements in their relationship. Without admonition, I think it is human nature to focus on the positive instead of the need to change our ways, and an admonition appears to be missing here.

This looks like a very slippery slope to me, whether it represents a change or not.

I think John Chapter 8 may be very instructive here, if the priest doing such a blessing might follow Jesus’ example (which should, of course, be a no-brainer). I added the bold to the most important part:

There is a reason the NIV has the whole thing in brackets; it’s extremely dubious. The NIV was not put together by a bunch of atheists with chips on their shoulders (and neither was the NRSV); if even they found it problematic, then…it’s problematic.

The earliest and most reliable manuscripts and other ancient witnesses do not have John 7:53-8.11

I think you’ve said elsewhere that it is quite difficult to determine what Exactly constitute the “original” scriptures…this potentially controversial passage, having been now included for many hundreds of years continues to require footnotes but, imo, it should still be included in current & future bibles, but continuing to note the controversy in footnotes in vital.

Actually my printing of the NIV does as much as can be expected. The warning is in the main text flow, where you can’t miss it. (The NRSV puts [[ ]] around it and has a footnote.)

There are many instances where it’s very difficult to pin down the original wording, and I agree it’s very important to note those.

But honestly, this is not one of them. There’s very little doubt, it’s about as ironclad as things get with ancient texts. It almost certainly wasn’t there originally.

Having done some reading this morning, I see what it is you are saying.

The story of the adulteress is a late entry. There is controversy around where it came from.

John is a completely “own its own” Gospel, different from the other three.

Each Gospel writer had a different perspective, and different goals for what they were writing. John doesn’t contain the story of water into wine, raising Lazarus, or parables. There wouldn’t need to be multiple Gospels if they all said exactly the same things! John appears much more focused on Jesus’ divinity. His first words place Jesus with God in the beginning, when he was the Word spoken to begin Creation.

We laugh in my church about the NIV, and call it the Nearly Inspired Version. I am partial to the New King James, myself.

Interpreting Scripture is an arduous thing. I am improving, but I have a long way to go. But to dismiss the story of the woman at the well because it doesn’t appear in other books doesn’t make sense to me. No one knows who wrote the book of John, not for certain. Who knows whether he had insight and/or information the others didn’t. If it was John the Apostle who wrote it (what most people think), then John worked in Samaria after Jesus’s death and resurrection (see Acts Chapter 8). He may have heard the story of the woman at the well from people there, or even from her.

I find the NIV is actually pretty good at identifying alternate readings and also calling out situations where ancient witnesses and manuscripts disagree with each other. (There are other complaints about it I am sympathetic with.) And yes I’ve heard “Nearly Inspired Version” too. Of course it won’t have the latest manuscript discoveries in it, but neither will anything else.

The reason I cited it in particular is I was responding to Valerie and I know she uses it.

For as long as I can remember, when I click on a yootoob video link, it takes me to a screenshot of the video.

As of maybe two weeks ago, when I click on a yootoob link, it takes me to a black screen. The rest of the page looks normal, I can see other links with screen shots down the right side of the screen, and the title under the video, the author’s info, the comments section, etc., but the video itself, where the screen shot or ‘title screen’ used to be, is now BLACK.

So if I want to see the opening screen of the video, I have to click play and then pause.

Does anyone know what has changed, and how I can change it back, so that when I click on a youtube link, it takes me to the screenshot / opening image of the video, instead of the black screen?

Whoa!!! Blue Lagoon in Iceland is a major tourist destination, people love to swim there.

That’s not an out-of-the-way location.

Almost everyone in Iceland lives very near the shoreline, the center of the country is a glacier that covers many volcanoes. Sometimes one under the ice erupts and it gets interesting.

There’s a show on Netflix called Katla, I think, that takes place in Iceland. I watched it in Icelandic w/ subtitles. What an unusual land & people. I think many of them are descendants of Vikings & their language is based on Scandinavian languages, to some degree, I believe.

It’s definitely a Scandinavian language, and it’s one that hasn’t changed much. They can still read the old sagas. They’re very close to what the vikings spoke (and yes, they descend from Viking settlers). Iceland is basically the only place on Earth where the original settlers are still in control–the island was uninhabited when the Vikings came.

It’s considered European…largely because it was settled by Europeans originally.

It is actually on the plate boundary, which is why they have so much volcanic activity. Reykjavik is actually on the North American plate. Most of the island is on the European plate.

Thingvellir park (which includes the first Althing or parliament building) is actually on the rift itself; it’s possible to scuba dive and touch both continents at once.

The driver was a woman. I didn’t expect that.

Police got the 911 call just after 10:15 p.m. They found five people at the scene with life-threatening injuries. All were transported to the hospital, and there has been no update on their conditions from officials as of Monday morning. 11 News has reached out to the police department for more information.

Based on the investigation so far, police say they believe the crowd met at the strip mall specifically to do donuts in the parking lot. The alleged driver of the SUV, who did not sustain serious injuries, was arrested for reckless driving. She has been identified by CSPD as Marisol Wentling. Oh, and I have trouble remembering what math class goes with what year of high school. 1) I went to one of those three year high schools with a junior high. So 9th grade wasn’t high school…except that legally it was, it counted toward your high school GPA. And I was also a year ahead; I got put into “Accelerated Math” in 7th grade, where they crammed 7th and 8th grade math into one year. So I had first year Algebra in 8th grade, geometry in 9th grade…and for my first year actually IN high school, tenth grade, I had algebra II. I have to remind myself that algebra I and geometry are high school classes, normally, even though I took them in junior high school. (My old school district has since gone back to “middle school” grades 6-8 and “normal” high school 9-12.)

Yeah, that was usual. Algebra I in JHS, Geometry et. seq. in HS. I just happened to be a year faster than most. So I ended up going through high school math classes (and science classes) with about half of the people in the class a year ahead of me. Until calculus, that is; those older kids were off to college by then.

I’m sure some people took algebra I in tenth grade or later because they needed one year of math to graduate.

I took physics my junior year (and again in college, much more intense). The one AP option for senior year was anatomy and physiology. Gooshy stuff, I wasn’t interested. So I took some off the wall stuff, geology and astronomy. The astronomy class was…long before I took it…the capstone class, physics as a prereq. When I took it, it was an “easy” option for someone needing their one year of high school science. There was an option to chase an honors A (5.0 on the GPA) and I took it. It still wasn’t hard. The teacher (poor guy–he was pursuing a doctorate and had to teach the easy class) had trouble explaining radians to the students and actually asked me one day after class if I had any ideas about how to do it. He used my suggestion and unfortunately it didn’t work.

My calc class was the period before physics and was sync so we got the calculus to derive the formulas before we got the spoon fed formulas. That made it easy to Ace physics.

And in point of fact, I was taking that class when I had physics…as a junior.

I noticed people struggled with related rates–that was the first big sticking point (the classic teaching problem there is writing a formula for how fast the water level changes as a conical tank, point down, drains). It’s sort of like everything that went before it…only you kind of back into it from the other side.

We never did conicals that I can recall. Derivatives & area under the curve & even logarithms were not really making sense to me at the time.

Your explanations in many of your science series posts do make Some sense to me, but my aging brain just doesn’t retain the info very well now. When reading your posts I refer back to the illustrations & written explanations numerous times but don’t sweat it all too much since I realize most info will subsequently drain out of my “cone” head anyways! LOL

I’ve always been a bit of a night-owl.

When I took engineering calculus in college, it was taught over three quarters. I got the highest A in the first quarter (derivatives) and even got a big push to switch to the mathematical calculus track for the second quarter.

Instead, I continued in the engineering track for integral calculus and ended-up taking a NoPass. My parents (five STEM degrees between them, one a professional engineer and the other a math professor….) were, naturally, appalled.

The first quarter was at two in the afternoon, the second quarter was at eight in the morning.

I only ever had single roommates in a dorm room too. As seniors there were excess females on campus so some of us got to live in Grad Housing apartments. I was in a 2 BR apt w/ 3 other girls but my roomie left at Christmas so my final semester I had that BR to myself.

It was actually a fairly weird experience having roommates since I was the only girl in my family & never had to share a room before…& coming from a more male-dominated household I really just didn’t get how chicks thought & definitely not how they emoted. It was a pretty bizarre & steep learning curve!

It’s a good interview. At about 20 minutes in, Tucker says America was founded on Western universalism, the idea that principles are to be universally applied and that they apply to all people equally. Tucker thinks we are now living under an Eastern hierarchical system where some people are deemed better than others. They think they deserve different treatment, and they live by different standards.

IMO this helps to explain why they hate the Constitution and America so much. It’s globalism, and it’s also that they’re trying to steer us away from the standards of Christian, Western civilization and America’s founding. (Tucker says you don’t have to be a Christian to benefit from living in a Christian society.) They can’t be above everyone else if everyone is equal. This is a profound battle.
